On 18 May 1749 when Samuel was 28, he first married Comfort Learned (31271) , daughter of Ebenezer Learned (31 Aug 1690-15 Mar 1772) & Deborah Haynes (13990) (30 Jul 1690-21 Aug 1777), in Oxford, MA. Born on 11 Jul 1730 in Oxford, MA. Comfort died in Worcester, MA, on 11 May 1765; she was 34.

On 27 Feb 1767 when Samuel was 46, he second married Sarah Trask in Beverly, MA. Born ca 1725. Sarah died in Worcester, MA on 10 Oct 1774.

On 8 Jan 1776 when Samuel was 55, he third married Esther Kittredge (39378) , daughter of James Kittredge (22 Aug 1687-23 Jan 1754) & Sarah Fowle (20004) (17 Aug 1696-aft 19 Jan 1754), in Lancaster, MA. Born on 26 Feb 1736 in Tewksbury, MA. Esther died in Northboro, MA, on 1 Aug 1815; she was 79.

Esther first married Josiah Locke, second Samuel Mower as his third wife.
